Cagrilintide

A long-acting amylin analogue, developed both alone and combined with semaglutide as CagriSema. Amylin acts on a satiety pathway distinct from the incretin axis.

Mechanism

Amylin is co-secreted with insulin by beta cells and signals satiety through the area postrema. Because the pathway differs from GLP-1, combining the two produces additive appetite suppression rather than overlapping effects.

Clinical evidence

REDEFINE-1 reported mean weight reduction of about 22.7% at 68 weeks for CagriSema in adults with obesity and without diabetes.
