Teduglutide

A GLP-2 analogue approved for short bowel syndrome. It is not a weight-loss drug — GLP-2 acts on intestinal growth and absorption, not appetite.

Mechanism

GLP-2 is cleaved from the same proglucagon precursor as GLP-1 but signals through its own receptor on subepithelial myofibroblasts and enteric neurons. Activation increases villus height, crypt depth and mucosal blood flow, improving absorptive capacity.

Teduglutide substitutes alanine with glycine at position 2 to resist DPP-4, extending the half-life from minutes to hours.
