A mitochondria-encoded peptide that acts as a metabolic regulator, activating AMPK and improving insulin sensitivity and exercise capacity in mice. Human data are observational rather than interventional.

Mechanism

MOTS-c influences folate-methionine metabolism and translocates to the nucleus during metabolic stress, regulating adaptive gene programmes. In mice it prevents diet-induced obesity and insulin resistance.

Human data

Circulating MOTS-c levels correlate with metabolic health and exercise adaptation, but no randomised human dosing trial has reported efficacy.
