Dulaglutide

A weekly GLP-1 agonist built as a fusion protein rather than a lipidated peptide. Approved for type 2 diabetes with a cardiovascular indication.
Mechanism
Two GLP-1 analogue chains are covalently linked to a modified human IgG4 Fc fragment. The large fusion construct resists renal clearance and DPP-4 cleavage, giving weekly dosing without fatty-acid albumin binding.
Clinical evidence
REWIND showed a reduction in major adverse cardiovascular events over a median 5.4 years in a largely primary-prevention population. Weight effects are modest compared with semaglutide or tirzepatide.
