Liraglutide
ALSO KNOWN AS · Victoza · Saxenda
The first-generation once-daily GLP-1 agonist, now largely superseded by weekly agents but still widely used and available as a generic in several markets.
Mechanism
Liraglutide is 97% homologous to native GLP-1 with a C16 palmitoyl chain that supports albumin binding and self-association at the injection site, extending exposure to roughly a day.
Clinical evidence
SCALE reported a mean 8.0% weight reduction at 56 weeks on 3.0 mg. LEADER demonstrated a reduction in cardiovascular death in type 2 diabetes, the first such result for the class.
