The other incretin hormone. Long dismissed as therapeutically inert in diabetes, GIP became central to the class once dual agonists showed it improves both efficacy and tolerability.
The reversal
GIP's insulinotropic effect is blunted in type 2 diabetes, which is why single-agent GIP was abandoned for decades. Restoring glycaemic control appears to resensitise the pathway, and tirzepatide's results forced a re-evaluation of the hormone's role in adipose tissue and central appetite regulation.
The field remains split on whether GIP receptor agonism or antagonism is the better strategy — both are in clinical development.
