Oxyntomodulin
A naturally occurring proglucagon product that activates both the GLP-1 and glucagon receptors — the biological template for the modern dual and triple agonists.
Significance
Oxyntomodulin demonstrated in humans that combined GLP-1 and glucagon signalling reduces food intake while raising energy expenditure. Its weak potency and short half-life made it unusable as a drug, but its receptor profile is the direct ancestor of survodutide, mazdutide and retatrutide.
