Why most peptides have to be injected
Absorption, not chemistry, is what keeps peptides out of pill form — and what a few programmes have done about it.

The gut is a protein shredder
Peptides are short proteins. Swallow one and gastric acid plus pancreatic proteases dismantle it before it reaches circulation. Even if it survives, the intestinal wall barely lets molecules of that size through — oral bioavailability is often well under one percent.
Three ways around it
Absorption enhancers: oral semaglutide is co-formulated with SNAC, which raises local pH and helps the peptide cross the gastric wall. Small-molecule mimicry: orforglipron is not a peptide at all but a small molecule that activates the same receptor, so it behaves like an ordinary tablet. Alternative routes: intranasal delivery, used for several neuropeptides, bypasses the gut entirely.
